Labelling and packaging of samples

In order to ensure the safe transmission of your samples the labelling and packaging use:

-Use small micro centrifuge tubes (0.5-0.65 ml)
-Please use simple labels on your tubes (e.g. initials and numbers)
-Write the label clearly on the side and top of tube with a permanent marker!
-Seal the lid with a small piece of parafilm
-Place samples in polybags, bubble wrap and send in padded envelopes

Results distribution

Sequencing results are sent to you via e-mail, the prints via normal post. Any further questions can be emailed to sequencing@fusionantibodies.com.

Sequencing Costs

The cost of automated sequencing is.

-Single sequence read* : £10.00
-Same day sequence read**: £15.00

*Discounted rates are available for large scale sequencing projects.

Fusion Antibodies has put together the following guidelines to enable the successful sequencing of your templates.

Automated DNA Sequencing Guidelines

For automated DNA sequencing you need to supply purified DNA and sequencing primer. We have various standard primers available and we can have them made quickly if desired, enquire for further details.

It is important that the DNA template and primer be as free of salt as possible. The quality of the sequencing and the length of read is very dependent on the quality of the starting DNA, more so than with manual sequencing. For purifying plasmid or PCR products we strongly recommend that you use only those kits or methods that carry an application test for Fluorescent DNA Sequencing. These products are specifically developed and optimized with automated fluorescent sequencing in mind. They are very reliable and usually provide templates that produce clean and long sequences. With high quality DNA and primer, the length of sequence read can get as high as 900 bp with > 98.5% accuracy. We can also purify your DNA for you if preferred.


Primers
We have a range of the most commonly used primers for sequencing, including M13 forward and reverse primers and the T7 forward primer.

Your sequencing primer should have a GC content of 50%, and have a Tm between 48 and 65 ºC. Best results will be achieved with a Tm of 55 ºC. Please indicate your primer sequence on the order form. You need to supply per template a minimum of 5 µl of the sequencing primer @ 3.2 pmol/µl.

It is essential that concentration of DNA be accurate. Please adjust concentration according to the specifications below. Samples not complying will be delayed considerably in processing.

Plasmid
If you are sequencing plasmid DNA you need to supply for each reaction a minimum of:
5 µl of double stranded DNA @ 250 ng/µl
5 µl of single stranded DNA @ 75 ng/µl
GC Rich template or suspected secondary structure (plasmid) DNA (i.e. 60-65%): Per reaction a minimum of: 5 µl of DNA @ 1 µg/µl.


Cosmid/Lambda
Per reaction a minimum of: 5 µl of DNA @ 2 µg/µl.


PCR Products
Per reaction a minimum of:
5 µl for PCR product size ranges from 200-1000 bp @ 20 ng/µl
5 µl for PCR products size range from 1001-3000 bp @ 100 ng/µl
If >3000 bp treat as plasmid DNA
If you suspect your PCR product is GC-rich or has secondary structure, please use double these amounts and let us know in your comments on the order form so that the correct protocols and thermal cycling programs can be used.
